What is Nitric Oxide?
In 1998, the Nobel Prize in Physiology or Medicine was awarded to American scientists Ignarro, Furchgott and Murad. They discovered and described a gas that is produced and released in blood vessel walls and regulates vessel function: Nitric Oxide, or NO for short. To better understand the effect of NO in the body, one need only think of the famous blue pills for improved performance, which inhibit NO breakdown and thereby stimulate blood circulation. Heart medications work in a similar way, which are based on organic nitrates or nitro-glycerin. Here too, the strongly released Nitric Oxide is intended to dilate the arteries and promote blood circulation.

Nitric Oxide is produced in the endothelial layer of blood vessels and causes dilation or relaxation of smooth vessel muscle. Medical science has published impressive results on NO over the past 10–20 years. Thus, Nitric Oxide generally promotes vessel health – moreover, a good NO level is a prerequisite for blood vessels to remain healthy at all. A fall in NO levels, which is typically found in the context of Western, that is to say modern lifestyles, causes blood vessels to become dysfunctional, referred to as endothelial or vascular dysfunction (Cyr et al. 2020). Long-term consequences include arteriosclerosis or other vascular diseases.
In recent years, it has additionally been discovered that Nitric Oxide has very, very far-reaching effects on the body. Thus, it regulates the function of the immune system, and moreover that of cellular energy metabolism. That is, vessel health and general health and in particular the health of energy metabolism are closely linked to one another.
Nitric Oxide (NO) is a signalling molecule that is synthesised in animals by NO synthase from L-Arginine. There is increasing evidence that NO regulates the metabolism of energy substrates in mammals.
NO discoverer and Nobel laureate Louis Ignarro wrote the book „NO More Heart Disease: How Nitric Oxide Can Prevent--Even Reverse--Heart Disease and Strokes" based on these findings and his research. Because: NO can be positively influenced by lifestyle factors. Thus, the starting substance for the formation of NO is the amino acid Arginine.
Why Citrulline?
Arginine is, however, rapidly metabolised with supplementation. Citrulline, on the other hand, bypasses these effects and increases the arginine level in the blood more strongly than arginine itself. Generally speaking, some substances that can be ingested through food have a positive influence on the modulation of NO levels.
L-Citrulline supplementation led to an increase in arginine flux and to a stronger increase in arginine concentration in the plasma than arginine supplementation itself. Whilst citrulline escaped first-pass metabolism, 70% of the supplemented arginine disappeared before it reached the peripheral circulation.
